ミトコンドリア、ゴルジ体に関連する応答ゾーン、連携ゾーン解析
研究概要
本研究では、「ミトコンドリアとゴルジ体の応答ゾーン」、「ミトコンドリアとゴルジ体の連携ゾーン」を中心に解析する。
私達は、細胞の生存や増殖、分化に関わるミトコンドリア応答やゴルジ体でのストレス応答が、オルガネラ膜上の限局された「応答ゾーン」での素反応の集積であることを見出した。また、生体応答の様々な場面でミトコンドリアと小胞体、あるいはミトコンドリアとゴルジ体の間での直接接触(連携ゾーン)を介した物質交換が行われていることを見出した。本研究では、これらの「応答ゾーン」、「連携ゾーン」を解析することにより、オルガネラ動態や役割を正確に解明する。具体的には、細胞死の実行時やカルシウム過負荷の時に生じる「ミトコンドリア応答ゾーン」や、ゴルジ体ストレス時に生じる「ゴルジ体応答ゾーン」に関して、①応答ゾーンの動態観察、②応答ゾーン形成に関わる分子の同定、③応答ゾーン形成機構の解明、④応答ゾーンの生物学的役割の解明を行う。
また、ゴルジ体ストレス時に生じる「ミトコンドリア⇔ゴルジ体連携ゾーン」に関して、①連携ゾーンの動態観察、②連携ゾーン形成に関わる分子の同定、③連携ゾーン形成機構の解明、④連携ゾーンの生物学的役割の解明を行う。さらに、人為的にオルガネラ連携ゾーンをon/offできる技術を開発し、連携ゾーンの機能解明に応用する。
