研究概要
細胞内外からの様々なストレスは、小胞体不良タンパク質を蓄積させ、小胞体さらには細胞の機能障害をもたらす。このような状況を打破するために、細胞はunfolded protein response(UPR)により小胞体ホメオスタシスの維持を図る。私たちは、小胞体に関連した次の2点について研究を進める。
<小胞体分解ゾーンに関する研究>
小胞体タンパク質の分解としては、内腔および膜上の不良タンパク質分解システムER-associated degradation(ERAD)に加えて、タンパク質を小胞体内腔へ挿入する前に、翻訳後すぐに細胞質で直接分解し小胞体負荷を軽減させるシステムER stress-induced preemptive quality control(ERpQC:予防的品質管理)が存在する(Cell Rep. 2015)。これまでに、ERADとERpQCの構成因子として、両者に共通する分子(Derlinやp97)とともに、共通しない分子(Sec61αなど)の存在を見出してきた。これらの知見をもとに、ERADとERpQCのゾーンが異なることを明らかにすることで、小胞体膜上の分解について異なる機能場が存在すること明らかにしたい。
<ミトコンドリア-小胞体連携ゾーンに関する研究>
UPRセンサー分子の一つは、小胞体ストレスだけではなくミトコンドリアストレスにも応答し、特異的リン酸化を介したストレス応答に重要な役割を担うことをこれまでに発見している。そこで、ミトコンドリア-小胞体連携ゾーンにおける小胞体センサーを介したストレス応答の分子メカニズムと生物学的意義の解明を目指す。
