小胞体膜連携ゾーンを介した脂質輸送
研究概要
小胞体で生合成された各種の膜脂質はそれぞれに異なるオルガネラへと移動していますが、どのようにしてそのような移動が起こるのかは長い間不明でした。しかし、小胞体で合成されたセラミドをスフィンゴミエリン(SM)に変換させるゴルジ体領域へと選別輸送する分子マシーンCERTを私たちが発見したことを契機として、その作動原理が明らかになってきました。すなわち、異なるオルガネラが極近距離に配置された細胞内亜領域(膜接触部位:membrane contact sites, MCS)においてCERTのような脂質転移タンパク質(lipid transfer proteins; LTPs)が効率的にオルガネラ間の脂質輸送を実行していることが様々な生物種、多様な脂質種で広く明らかになってきたのです。しかし、オルガネラゾーンの一種であるMCSの形成と制御の詳細な機序は今でも不明のままです。そこで、本領域研究において私たちは以下のことを目指します。小胞体-ゴルジ体連携ゾーン形成に関与する遺伝子を幅広く同定し、それら遺伝子の果たす役割を明らかにします。また、CERTの機能をコンロトールする機序を明らかにします。これらの解析においては分担者の加藤が得意とする超解像度顕微鏡観察を駆使し、さらに、植物細胞における脂質輸送を研究する分担者の下嶋とも連携して、動植物の間に共通するような脂質輸送の作動や制御の仕組みを明らかにすることも目指します。水に溶けない低分子化合物である脂質を細胞内のような水環境下において的確に移動させている生命体の驚異的な働きの全体像が本計画班の研究から浮かび上がってくると期待されます。
