Visit of Dr. Keisuke Ito from Albert Einstein College of Medicine, USA

On May 10, 2023, Dr. Keisuke Ito, Associate Professor at Albert Einstein College of Medicine visited our laboratory. He gave us an inspiring and meaningful seminar on hematopoietic stem cells, particularly in view of single-cell fate decision-making and mitochondrial regulation.

Visit of Dr. Nattawat Onlamoon from Mahidol University, Thailand

Assoc. Prof. Dr. Nattawat Onlamoon, Research Supervisor of Tarapongpun Tanakorn (Joint Degree Program Student) at Mahidol University visited us on April 24, 2023. We had a good discussion on scientific issues of mutual interest particularly related to Tanakorn Tarapongpun's project.

Organization of self-advantageous niche by neural stem/progenitor cells during development via autocrine VEGF-A under hypoxia.

We show that neural stem/progenitor cells (NSPCs) secret VEGF-A in an autocrine fashion to efficiently maintain themselves under hypoxic developmental environment. Our results suggest that NSPCs have adaptive potential to respond to hypoxia to organize self-advantageous niche when the vascular system is immature.

We newly found a mechanism of glioblastoma recurrence by which cancer cells in brain replenish oxygen and iron for the self-expansion of cancer stem cells through systemically promoting host erythropoiesis.

We review the mechanisms of glioma progression and recurrence by focusing on how GSCs organize their self-advantageous niche environments, which were revealed by Tabu et al.'s studies with a unique approach using polymer microarrays screening and with a unique viewpoint of necrotic particles.

Sox17-mediated expression of adherent molecules is required for the maintenance of undifferentiated hematopoietic cluster formation in midgestation mouse embryos.

Hematopoietic stem cell-containing intra-aortic hematopoietic cell clusters emerge in the dorsal aorta at midgestation. A transcription factor Sox17 directly induces the gene expression of adherent molecules, VE-cad and ESAM. These adherent molecules have an important role in the formation of hematopoietic cell clusters and the maintenance of undifferentiated state at least in the coculture with stromal cells.

Students of Toyama High School visited the Department of Stem Cell Regulation on August 21, 2019. They received mini-lectures from the Professor. The students observed fluorescently labeled cancer stem cells attached to synthetic polymers. They also experienced the mouse embryo observation, the microscopic (phase contrast and fluorescent) observation of undifferentiated hematopoietic cells, the harvesting of the cells, the immunofluorescent staining, and the identification and sorting of hematopoietic cells.

Medical students of TMDU's partner universities in the United States, Thailand, and Taiwan participating in TMDU’s 8th International Summer Program (ISP2019) visited the Department of Stem Cell Regulation on July 9 and 10, 2019. They received mini-lectures from the Professor. The students observed fluorescently labeled cancer stem cells attached to synthetic polymers. Students of Osaka Ibaraki High School visited the Department of Stem Cell Regulation on August 7, 2018. They received lectures by the staff members of the department, followed by practices of microscopic observation of cultured cells including stem cells.

Students of Senior High School at Otsuka, University of Tsukuba, visited the Department of Stem Cell Regulation on July 23, 2018. They received mini-lectures from the Professor and Assocoate Professor as well as a Technical Assistant of the department. The students experienced the microscopic (phase contrast and fluorescent) observation of undiffrentiated hematopoietic cells, the harvesting of the cells, the immunofluorescent staining, and the identification and sorting of hematopoietic cells.

Prof. Mauro Teixeira at Department of Biochemistry and Immunology, Universidade Federal de Minas Gerais (UFMG), Brazil visited the Department of Stem Cell Regulation and gave us a seminar entitled “- Inflammation and its resolution - mediators and relevance “ on June 22, 2018.

 

Visit of Prof. Chambers, Dr. Burdon and and their colleagues from the University of Edinburgh.

Dr. Ian Chambers, Dr. Tom Burdon, Dr. Zhang, and Dr. Ozkan from the University of Edinburgh visited the Department of Stem Cell Regulation on April 18, 2018. We discussed recent findings on hematopoietic and cancer stem cells made by the TMDU side and helpful comments were given by the Edinburgh attendees. Later, we enjoyed our short walk to Kanda Myojin and had a good time together with a dinner.

Maintenance of hematopoietic stem and progenitor cells in fetal intra-aortic hematopoietic clusters by the Sox17-Notch1-Hes1 axis. Experimental Cell Research, 365: 145-155, 2018.

 

In the mouse ontogeny, the dorsal aorta, from which definitive hematopoiesis firstly arises in midgestation mouse embryo, has hematopoietic stem cell-containing clusters. This paper show that the Sox17-Notch1-Hes1 pathway contributes to maintaining the undifferentiated state of the clusters.

Visit of students from Senior High School at Komaba, University of Tsukuba.

Students of Senior High School at Komaba, University of Tsukuba, visited the Department of Stem Cell Regulation on December 14, 2017. They received mini-lectures from the Professor and Assistant Professor as well as a PhD student of the department. After the mini-lectures, the students experienced the microscopic (phase contrast and fluorescent) observation of cancer cell lines, the harvesting and counting of cultured adherent cells, and the identification and sorting of cancer stem cells.

A student from Tokyo Institute of Technology have joined our laboratory under the internship program.

Since November 14, 2017, Mr. Mongkol Pongsuchart, a student of Prof. Shinae Kondo’s Lab. at Tokyo Institute of Technology (Tokyo Tech), joined us for one week-research under the Tokyo Tech Internship Program with the supervision of Dr. Tabu (Assistant Professor).

 

Visit of Dr. Ivan Velasco from Universidad Nacional Autonoma De Mexico.

Dr. Ivan Velasco from Universidad Nacional Autonoma De Mexico visited the Department of Stem Cell Regulation and held a seminar titled "Enhancing survival, maturation and axonal growth of stem cell-derived neurons" on Novbember 9, 2017. The presentation was quite insightful and many participants, including a TMDU student Erick Luz Madrigal, enjoyed discussion afterward. The conversation continued at dinner.

Visit of Prof. Bradley and Ms. Schmidt from the University of Edinburgh.

Our collaborators, Prof. Bradley and Ms. Schmidt, from the University of Edinburgh, Scotland, UK visited the Department of Stem Cell Regulation on August 25, 2017. We discussed recent advances and future prospects of our collaborative research on the elucidation of stem cell niche by using synthetic polymers.

Later, we enjoyed our short walks to Yushima Seido and Kanda Myojin and strolling in Sensoji as well. Amazake, a fermented rice drink, at Kanda Myojin was quite good at this time of year.

Thrombopoietin contributes to the formation and the maintenance of hematopoietic progenitor-containing cell clusters in the aorta-gonad-mesonephros region.

 

In the midgestation mouse embryo, hematopoietic cell clusters containing hematopoietic stem/progenitor cells arise in the aorta-gonad-mesonephros (AGM) region. This paper showed that thrombopoietin (TPO), a cytokine, contributes to the formation and the maintenance of hematopoietic cell clusters in the AGM region.

Enhancement of 5-aminolevulinic acid-based fluorescence detection of side population-defined glioma stem cells by iron chelation.

 

Cancer stem cells (CSCs) are dominantly responsible for tumor progression and chemo/radio-resistance, resulting in tumor recurrence. 5-aminolevulinic acid (ALA) is clinically used as a reagent for photodynamic diagnosis (PDD) and therapy of cancers including gliomas. We show that side population-defined C6 glioma CSCs (GSCs) possess an ability to escape from the PDD by displaying much less 5-ALA-derived fluorescence. We further show that the GSCs can be detected by 5-ALA in combination with deferoxamine, an iron chelator. Our results provide important new insights into 5-ALA-based PDD of gliomas.

The 20th anniversary party of Taga's Laboratory

It has been 20 years since Dr. Taga was appointed Professor at Tokyo Medical and Dental University (TMDU) on September 1, 1996. The 20th anniversary party of Taga's Laboratory was held in Yokohama on an occasion of the 39th Annual Meeting of Molecular Biology Society of Japan. (December 1, 2016)

Requirement of ABC transporter inhibition and Hoechst 33342 dye deprivation for the assessment of side population-defined C6 glioma stem cell metabolism using fluorescent probes. (November 4, 2016)

 

Elucidating the precise properties of cancer stem cells (CSCs) is indispensable for the development of effective therapies against tumors, because CSCs are key drivers of tumor development, metastasis and relapse. In this article, we show that inhibiting ABC transporters and depriving Hoechst 33342 dye are required for the accurate assessment of CSC metabolism, particularly side population-defined C6 glioma stem cell metabolism using fluorescent probes.

Young Investigator Papaer Award 2015 in Development, Growth & Differentiation.

Maha Anani, MD. PhD. has recieved the Young Investigator Paper Award 2015 of Development, Growth & Differentiation at 48th Annual Meeting of the Japanese Society of Developmental Biologists (June 4, 2015). Her award-winning achievements are as follows:

 

It has been reported that Sox17 is expressed in fetal hematopoietic progenitor cells in the aorta-gonad-mesonephros (AGM) region in embryos while Sox17 is involved in maintenance and differentiation of adult hematopoietic progenitors in bone marrow. Based on the information, this manuscript showed functions of Sox17 in fetal hematopoietic progenitor cells in the AGM by forced expression of Sox17 in those cells. The authors indicated that the Sox17 expressing progenitor cells preferentially developed to common myeloid progenitors (CMPs) after transplantation into irradiated recipient mice. In addition, Sox17-expressing progenitors cells gave rise to CMPs as well as granulocyte/macrophage progenitors (GMPs) in cultures, and their self-renewal proliferation was maintained for some passages. Moreover, those cells showed increased expression of key genes for hematopoietic differentiation compared with that in control cells without Sox17 transduction, and they formed the mixed type colonies consisting of multi-lineage hematopoietic cells in culture. In summary, this manuscript clearly demonstrated critical functions of Sox17 for maintenance and differentiation of fetal hematopoietic progenitors cells, in particular myeloid lineage, by detailed transplantation and culture experiments, and therefore, I would recommend this manuscript for the DGD award.

Three students acquired a master's degree in TMDU. Two are working at private companies and the one has entered the PhD course and is studying at another department.

A foreign researcher from Kon Kaen University in Thailand and an exchange student from Imperial College in UK have joined our laboratory

Mr. Pra-Yong Pokpong heve joined our laboratory for half a year as a foreign researcher from Kon Kaen University in Thailand. Ms. Georgina Phillips is participating in research by the schedule for about two months as an exchange student from Imperial College in the UK.

The manuscript entitled “CD45lowc-Kithigh cells have hematopoietic property in the mouse aorta-gonad-mesonephros region” has been published in Experimental Cell Research.

 

2011

The cancer stem cell niche project was reported in Nikkei Industrial Daily

We are search for the optimal microenvironment (niche), in which normal neural stem cells and cancer stem cells could maintain their self-renewal and pluripotential phenotype in vivo. We are screening artificial niche structures using chemically synthesized polymers in collaboration with Professor Mark Bradley and his colleagues in The University of Edinburgh. Our niche project concerning cancer stem cells was reported in Nikkei Industrial Daily on November 1, 2011.

Symposium on Understanding of Organogenesis Toward New Therapy was held

As a research meeting supported by Joint Usage/Research Program of Medical Research Institute, Tokyo Medical and Dental University, the symposium entitled Understanding of Organogenesis Toward New Therapy was held on October 21, 2011. The organizers were Prof. Shoen Kume of Institute of Molecular Embryology and Genetics, Kumamoto University, Profs. Hiroshi Nishina and the staff members of the Department of Stem Cell Regulation.

The best poster prize in 2011 workshop in young researchers for cancer

Dr. Kouichi Tabu(JSPS Fellow) has been awarded the best poster prize of 2011 workshop in young researchers for cancer (Aug. 31~Sep. 1. 2011, Artlandhotel Tateshina).

Visiting lectures at Chiba High School

The staff members of the Department of Stem Cell Regulation gave five lectures (40 students in each class) on stem cells, each followed by practice of microscopic observation of tissue specimens at Chiba Prefectural Chiba High School on June 17, 2011. This activity was held as a collaboration between Chiba High School and Tokyo Medical and Dental University.
