Anatomy and physiolgy in the genetically manipulated mice that have visualyzed longitudinal stripes in the cerebellum
Aldolase is an enzyme involved in one of the essential steps in catalyzing glucose. There are three isozymes, aldolases A, B and C, which are expressed predominantly in muscle and brain, liver, and brain, respectively. Among them, aldolase C is expressed in distinct subpopulations of cerebellar Purkinje cells that are arranged in longitudinal stripe-shaped compartments in the rodent and other mammalian cerebellar cortex. These stripes, we think, represent basic structure of the cerebellar cortex. To facilitate physiological and anatomical studies on this structure, we generated a knock-in mouse strain in which fluorescent protein Venus is expressed in place of aldolase C (Aldoc-Venus mouse).
Projection patterns of Purkinje cell axons and climbing fiber axons have been already shown for aldolase C stripes in the cerebellar cortex. Therefore, one can has knowledge of axonal projection patterns for any given areas in Aldoc-Venus mice without making tracer injections. In cerebellar sections of Aldoc-Venus mice, the striped pattern of aldolase C (Aldoc) is clearly observed without immunostaining. Therefore, we can analyse detailed striped expression pattern of aldolase C in these mice.
In Aldoc-Venus mice, the striped pattern is clearly visible during surgery under anaesthesia. So, we can record from identified stripes and inject tracers in indentified stripes to label mossy fibers retrogradely .
