Summary

As the education program for young researchers of A3 Foresight Program, I visited Prof. Yasuhito Yuasa’s laboratory (Department of Molecular Oncology, Tokyo Medical and Dental University) in Japan. The main purpose of this visit was to learn how to treat gastric cancer stem cells with anti-cancer drugs and how to make paraffin-embedded blocks of sphere cells. During my stay in TMDU, I could learn a lot of techniques and knowledge on analysis of cancer stem cell. I deeply thank Dr. Se-Jin Jang of Asan Medical Center and Prof. Yuasa to give me this chance and I hope that A3 foresight program will develop more and more.

1. Making of paraffin-embedded blocks of sphere cells

Recently we succeeded in cultivation of cancer stem cells to obtain spheres, but it was difficult to obtain light micrographs of the spheres. Thus I prepared spheres and fixed them with 95% EtOH in AMC, and brought them to TMDU. In TMDU, Dr. Fukamachi taught me how to prepare paraffin-embedded blocks and how to get serial sections using a rotary microtome. It was the first time for me to use a rotary microtome, but I found it necessary to use it to examine light micrographs of small samples.

2. Treatment of gastric cancer stem cells with anti-cancer drugs.

We found that cancer stem cells could be cultivated in a serum-free condition on some matrix. In treatment of cancer stem cells with anti-cancer drugs, we used MKN45 and MKN74cell lines, which could grow in the culture condition for cancer stem cells. could selected control cell line, which can growing in csc media and culture dish. In gastric cancer, Dr. Fukamachi used two kind of gastric cancer cell lines, MKN7 and MKN45 as control cell. After selection of control cell, On the first day, we seeded control and gastric cancer cells into wells of 24-well plates. On the next day, we treated the cells with various chemotherapeutic agents including 5-FU, DXF, CDDP and DXR. After 2 weeks, cell growth was determined by using MTT assay.
